A major fire has killed at least two people and injured others at the four-star Eurostars David hotel in the city center.
People are still stuck in the smoking building waiting to be evacuated, local reports said.
“There are two (deaths) but we fear that this is the final figure,” Petr Kolouch, director of the Prague Emergency Medical Service, told Czech television.
He said the smoke choked or poisoned the dead but they did not burn alive.
Others are now being treated for smoke inhalation.
Two firefighters are among those injured after they fell while dealing with the blaze, Lidovky.cz reports.

“It’s no longer burning, but the hotel is still very hot and smoky.
“We are still looking for people,” Prague Fire Department spokesman Martin Kavka told Lidovky.cz.
“We will be here for several more hours,” he added.

The cause of the fire is not yet known.
A firefighter told local media that the fire started on the ground floor and smoke had swept through the building.
At least 16 fire brigade units and a police helicopter are on the scene, it has been learned.

The Czech fire service said 40 people were evacuated from the hotel, located near the Vltava River and the National Theater in the Czech capital.
